Early diagnosis and prognosis of hepatocellular carcinoma based on a ceRNA array

Abstract Objectives Hepatocellular carcinoma (HCC) is one of the most common causes cancer-related deaths, due to high morbidity, a low early diagnosis rate, and poor prognosis. It essential explore competitive endogenous RNA (ceRNA) arrays for prognosis HCC. Methods The original gene expression profiles differentially expressed lncRNA, miRNA mRNA in HCC were downloaded from TCGA database. Differentially lncRNA-miRNA miRNA-mRNA interaction pairs extracted miRcode starBase, ceRNA network was constructed, GO annotation KEGG pathway analyses performed. Cox regression Kaplan-Meier survival analysis screening identified core genes associated with survival, centering on miRNA, which screened using arrays. qRT-PCR used detect level key clinical samples. Dual luciferase reporter assays verify target binding relationship among mRNA, miRNA. ROC curves analyze diagnostic efficacy array. Results A total 8 lncRNAs, 5 miRNAs, 21 mRNAs construct network. Functional enrichment showed that mainly enriched 14 signaling pathways, especially microRNAs cancer. Survival lncRNA FOXD2-AS1 miR-9-5p related HCC, targeted relationships between mRNAs. STMN1, COL15A1, CCNE2 TargetScan, miRDB, PicTar databases reliable. levels FOXD2-AS1, miR-9-5p, COL15A1 , upregulated tissues.
